Abstract: This paper presents a method for the preparation of alkaline phosphatase (EC 3.1.3.1) from calf intestinal mueose. The method involves extraction of the mucose with butanol, fractionation with acetone and then (NH4)2SO4, DEAE-cellulose ion-exchange column chromatography and Sephadex-gel filtration. The specific activity of the enzyme preparation was about 845 units mg of protein. The recovery of activity was 29%. Only one activity band was shown by the polyacrylamide gel electrophoresis.
